2X Castenholz TYE Media (makes 2L) - Mix the following ingredients:

2X Casteholz Salts (makes 1L) - Combine the following ingredients:
  • 0.2 g Nitrolotriacetic Acid
  • 2.0 mL Nitsch's Trace Elements (Link to this recipe)
  • 2.0 mL FeCl3 Solution (0.03%)
  • 0.12 g CaSO4-2H2O
  • 0.2 g MgSO4-7H2O
  • 0.016 g NaCL
  • 0.21 g KNO3
  • 1.4 g NaNO3
  • 0.22 g Na2HPO4-7H2O
  • Add distilled water until the final volume is 1.0 L.
  • Adjust the PH to 8 using 2M HCL and 2M NaOH


Nitsch's Trace Elements (makes 1L) - Combine the following ingredients:
  • 0.5 mL H2SO4
  • 2.2 g MnSO4
  • 0.5 g ZnSO4
  • 0.5 g H3BO3
  • 0.016 g CuSO4
  • 0.025 g Na2MoO4
  • 0.046 g CoCI2-6H2O
  • Add distilled water until the final volume is 1.0 L.


1% TYE (makes 200mL) - Combine:
  • 2.0 g Tryptone (difco 0123)
  • 2.0 g Yeast Extract
  • Add distilled water until the final volume is 200mL


The next step of our process is DNA isolation. During this step, we isolate the DNA in our samples for use in PCR.
